      Background InformationChromeobox 4 (CBX4), also known as Pc2, is a polycomb group (PcG) protein that functions as a transcriptional suppressor. CBX4 is thought to act as a co-repressor of DNMT3B, inducing transcriptional repression independent of DNA methylation. CBX4 also acts as a SUMO E3 ligase, facilitating SUMO1 conjugation by UBE2I. CBX4 is phosphorylated on Thr495 by HIPK2 upon DNA damage, which enhances its E3 SUMO-protein ligase activity.
