      Background InformationSUMO conjugation to target proteins is mediated by a different, but analogous, pathway to ubiquitinylation. UbcH9 is a SUMO conjugating enzyme that mediates the conjugation of SUMO-1 to a number of proteins including RanGAP1, IκBα and PML. It is known to interact directly with proteins that are modified by SUMOylation and may be important for substrate recognition. It appears that conjugation of SUMO-1 to IκBβ prevents subsequent ubiquitinylation and thus protects it from proteasomal degradation. Thus, while ubiquitinylation targets proteins for rapid degradation, SUMO-1 modification acts antagonistically to generate proteins resistant to degradation.

Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This gene encodes a member of the E2 ubiquitin-conjugating enzyme family. The encoded protein shares 100% sequence identity with the mouse and rat counterparts, which indicates that this enzyme is highly conserved in eukaryotes. Two alternatively spliced transcript variants encoding the same protein have been found for this gene.
