The Guava® easyCyte systems use patented, microcapillary, laser-based technology capable of detecting mammalian and microbial cells and beads. A sample of fluorescently labeled cells is aspirated into a uniquely proportioned microcapillary flow cell. Forward and side scatter characteristics are detected by photodiode, and fluorophores excited by the violet, blue, or red laser emit signals that are spectrally filtered to resolve up to 10 fluorophores simultaneously.
Accuracy on the Guava® easyCyte 8HT is shown using a human whole blood cocktail (FlowCellectTM Human CD4/CD8 T Cell Kit). Results were comparable to a predicate method on a sheath-fluid based flow cytometer.
Accuracy on the easyCyte™ 8HT is comparable to traditional flow cytometry
Superior small particle bead detection down to 0.2 μm now on the Guava® easyCyte 12 instruments.
Consistent Sample Results Across an Entire Plate with Sample Mixing
To ensure that cells stay in suspension during sampling, a rotating, variable- speed mixing paddle with auto-cleaning is used between each well so there is no need for manual resuspension after loading the plate.
Evaluation of well-to-well consistency with sample mixing for a 96-well plate. Sample concentrations were within acceptable range for all samples within the plate. easyCheck beads were diluted at 1.0x10e6 and placed into all wells of a 96 well plate. The plate was then spun at 150xg for 5 minutes to mimic settling. Each well was acquired and concentrations were determined. Data show the concentrations for each well (solid line) as well as +/- 10% values (dotted lines)
The mixing feature virtually eliminates carryover between samples and mixes more efficiently than shaking plate holder formats. Data shown below confirms that carryover is minimized with this format
Consistent Cell Counts for Each Channel
Fluorescence resolution for all channels show consistent counts and population percentages, for whole blood
Scatter Resolution for 8-Color Staining
% of Population
488 nm Laser
635 nm Laser
Immunological Phenotyping 6-Color Assay
The Guava® easyCyte 8HT system accurately discriminates six different fluorochromes within a single assay. As shown in the data below, the percentages and absolutely counts of six different populations were evaluated from whole blood.
5 Decade Dynamic Range Enables Simultaneous Detection of Bright and Dim Fluorochromes
HEK-293 cells transfected with GFP (green) or untransfected (grey).
High calibration mode enhances the detection of very dim fluorochromes, as it improves the signal to noise ratio, even in the red and far-red (APC) channel.
Positive and negative populations are detectable in the same scale, even for ultrabright fluorochrome dyes or fluorescent proteins, as shown here.
Software Templates for Simple Acquisition and Analysis
InCyte™ templates are new applications-specific modules with unique features that can be customized to your preferences for analysis. These modules are available for new and existing FlowCellect™ applications.
Pre-Calibrated Instruments that Need No Adjustment
Every Guava® easyCyte instrument manufactured now has optimized calibrated performance to ensure instrument to instrument reproducibility and performance.
No user adjustments are necessary, as laser and optical filter alignments are fixed with buffers in place to reduce the effects of vibration transference from internal mechanisms or external sources.
Transfer Methods and Setting Between Any Guava® easyCyte, Even for Systems with Different Optical Layouts
The ability to transfer instrument settings and methods between instruments allows comparisons of data between instruments and cross-site validation studies.
Fully-adjustable instrument settings on Guava® easyCyte flow cytometers and GuavaSoft™ software allow the user to optimize, save, and retrieve acquisition settings for SSc and all fluorescent channels (except FSc and NIR2, available late 2012) and display templates.

Quality Control of Daily Performance
The guava easyCheck kit is used to verify system performance and maintain daily quality control before running any assays, and is superior to instrument calibration for this purpose. The kit consists of Easy Check standardized fluorescent beads and diluent, facilitating assessment of counting accuracy and fluorescence detection. The guava Easy Check kit is included with all instruments to enable daily, routine signal optimization and quality control.
