Clarification Made Easy


The current evolution to high cell density and high product titer cell cultures is placing a larger burden on traditional downstream clarification and purification operations. With high solid content and an increased number of submicron particles, separation techniques to remove all solids can be complex. To alleviate these bottlenecks, various types of pre-treatment are being employed to make the clarification process more efficient.

Polycationic polymer modified poly (allyl amine) [mPAA] and poly (diallyldimethyl-ammonium chloride) [pDADMAC] are very effective flocculation polymers that can be used as a pre-treatment solution when used in combination with our Clarisolve® depth filters. Such polymers can reduce the area required for depth filtration by at least a factor 2-3x. This results in consumable, water flushing, and manufacturing footprint savings, thereby providing an alternative to centrifugation.

Clarisolve® mPAA Flocculation Polymer
The Clarisolve® mPAA polymer is a stimulus-responsive flocculation polymer designed specifically to enable a simpler clarification of high density cell culture monoclonal antibody (mAb) processes. The Clarisolve® mPAA polymer helps minimize risk by providing low residual polymer levels with no negative impact on downstream unit operations, and is less dependent on the dosage of the polymer which increases robustness and results in a wide-dosing window.

Optimal flocculation results are delivered when used in a concentration of 0.1% and 0.5%. The stimulus-responsive nature of the polymer means lower residual polymer levels.

pDADMAC Flocculation Polymers
pDADMAC is a cationic polymer that is widely used as a flocculant in waste water treatment. We have developed a high-quality production and purification process suitable for use in GMP biotherapeutic manufacturing.

When used in a concentration between 0.01 and 0.05%, the pDADMAC polymer rapidly flocculates the negatively charged cells and cellular debris into larger particles via an ionic interaction mechanism.
