Background InformationDickkopf-related protein 2 (DKK2) is a member of the dickkopf family and are a necessary component of vertebrate development through its interactions with the Wnt signaling pathway. DKK2 has the ability to act as an agonist or antagonist of Wnt/beta-catenin signaling which depends upon the cellular context and presence of the co-factor kremen 2. DKK2 is expressed in the brain, skeletal muscle, lung, and heart. Mutations in DKK2 are associated with bone disease.

      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a protein that is a member of the dickkopf family. The secreted protein contains two cysteine rich regions and is involved in embryonic development through its interactions with the Wnt signaling pathway. It can act as either an agonist or antagonist of Wnt/beta-catenin signaling, depending on the cellular context and the presence of the co-factor kremen 2. Activity of this protein is also modulated by binding to the Wnt co-receptor LDL-receptor related protein 6 (LRP6). [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer's disease (By similarity).

SUBUNIT STRUCTURE: Interacts with LRP5 and LRP6 (By similarity).

      SUBCELLULAR LOCATION: Secreted.

TISSUE SPECIFICTY: Expressed in heart, brain, skeletal muscle and lung.

DOMAIN: The C-terminal cysteine-rich domain mediates interaction with LRP5 and LRP6 (By similarity).

PTM: May be proteolytically processed by a furin-like protease.

SEQUENCE SIMILARITIES: Belongs to the dickkopf family.
