Background InformationNLR family member X1 (NLRX1) belongs to the Nod-like receptor (NLR) family of proteins, a group of proteins characterized by their highly conserved nucleotide binding domain (NBD) and their leucine-rich-repeats (LRR). NLRX1 is the only member of this family localized to mitochondria, where it plays a role in antiviral signaling by blocking virus-induced interaction of RLH-MAVS, serving as a negative regulator of the antiviral responses mediated by MAVS. It also serves to up-regulate NF-Kappa-B and JNK-dependent signaling cascades as its overexpression induces reactive oxygen species (ROS). NLRX1 has been observed in high quantities within the heart, mammary gland, and muscle tissues. It has also been observed at the protein level in various cell lines including; JURKAT, HeLa, HL60, RAJI, THP-1, and 293T.

      UniProt Number
UniProt SummaryFUNCTION: Participates in antiviral signaling. Acts as a negative regulator of MAVS-mediated antiviral responses, through the inhibition of the virus-induced RLH (RIG-like helicase)-MAVS interaction. Has no inhibitory function on NF-Kappa-B and type 1 interferon signaling pathways, but enhances NF-Kappa-B and JUN N-terminal kinase dependent signaling through the production of reactive oxygen species.

      SUBUNIT STRUCTURE: Interacts with MAVS.

      SUBCELLULAR LOCATION: Mitochondrion outer membrane

TISSUE SPECIFICTY: Ubiquitously expressed. Strongest expression in mammary gland, heart and muscle. Detected in HELA, 293T, THP-1, HL60, RAJI, and JURKAT cell lines (at protein level).

SEQUENCE SIMILARITIES: Belongs to the NLRP family.

      Contains 4 LRR (leucine-rich) repeats.

      Contains 1 NACHT domain.

SEQUENCCE CAUTION: The sequence BAB15075.1 differs from that shown. Reason: Erroneous initiation.

The sequence BAG51714.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.

The sequence BAG51714.1 differs from that shown. Reason: Erroneous termination at position 951. Translated as Trp.
      Molecular Weight ~115/112 kDa observed.
Doublet observed may be due to incomplete maturation by mitochondrial processing peptidases (MPPs). MPP maturation is common process for many proteins of the mitochondrial matrix and some proteins of the inner membrane space (Arnoult, D., et al. (2009). Journal of Cell Science 122:3161-3168).
