Background InformationPHD3 (Prolyl hydroxylase domain-containing protein 3), also known as Hypoxia-inducible factor prolyl hydroxylase 3 (HIF-PH3 or HPH-1), is one of 4 PHD (PHD1-4) proteins which function as prolyl hydroxylases to effect oxygen homeostasis via hypoxia-inducible factor (HIF). This transcriptional complex is composed of an alpha-beta heterodimer that mediates a broad range of cellular and systemic responses to hypoxia. The PHD proteins hydroxylate HIF-1 alpha at 'Pro-402' and 'Pro-564', and HIF-2 alpha. PHD3, is expressed in response to hypoxia and causes apoptosis in oxygenated conditions in neural cells. When oxygen levels are normal, PHDs catalyze the hydroxylation of prolyl residues on HIF-1alpha. This post-translational modification targets this protein for proteasomal degradation via the von Hippel-Lindau ubiquitination complex.

UniProt SummaryFUNCTION: Catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates HIF-1 alpha at 'Pro-564', and HIF-2 alpha. Functions as a cellular oxygen sensor and, under normoxic conditions, targets HIF through the hydroxylation for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. May play a role in cell growth regulation in muscle cells and in apoptosis in neuronal tissue. Promotes cell death through a caspase-dependent mechanism By similarity.

      CATALYTIC ACTIVITY: An HIF alpha chain L-proline + 2-oxoglutarate + O2 = An HIF alpha chain trans-4-hydroxy-L-proline + succinate + CO2.

COFACTOR: Binds 1 Fe2+ ion per subunit.

      Ascorbate.

ENZYME REGULATION: Activated in cardiovascular cells and HelA cells following exposure to hypoxia.

SUBUNIT STRUCTURE: Interacts with MORG1 By similarity.

      SUBCELLULAR LOCATION: Cytoplasm. Nucleus.

TISSUE SPECIFICITY: Widely expressed at low levels. Expressed at higher levels in heart (cardiac myocytes, aortic endothelial cells and coronary artery smooth muscle) and placenta.

SEQUENCE SIMILARITIES: Contains 1 PKHD (prolyl/lysyl hydroxylase) domain.
      Molecular Weight ~ 27 kDa
