      OverviewThis unique formulation of cell culture freezing media is for cryopreservation of a broad spectrum of mammalian cells (see viability data). Freezing Media contain 10% of the active cryopreservation ingredient (either DMSO or Glycerol). The remaining 90% is a proprietary mixture of Calf and Fetal Bovine Serum. The reagents used in these products are certified to be virus and mycoplasma free.

      The sera utilized in this medium is approved by the United States Department of Agriculture (USDA) to ensure that sera is free of diseases such as Bovine Spongiform Encephalopathy (BSE).

      Application NotesPicking and freezing of ES cells:

      Wash Plate with PBS. Pick colonies into 96 wll plate (U shaped wells) with 35 µl of trypsin / well. Pick roughly 48 colonies, incubate at 37 °C for 10 minutes. Use Multichannel pipet to break up colonies. Transfer directly into 96 well plate with feeders and 150 µl of ES medium (without selection drug). Feed every day with ES medium containing selection drug (150 µg /ml neomycin, or 50 µg /ml of hygromycin). After 2 days cells should be ready to be frozen. Wash with PBS add 35 µl of Trypsin, incubate at 37 °C for 10 minutes, add 65 µl of ES medium. Use multichannel pipet to disperse cells. Transfer 65 µl into replica plate (U shaped wells) containing 65 µl of cold 2 x ES freezing medium. Wrap with parafilm, place on dry ice for 20 minutes, and transfer to -80 °C.
