      OverviewThe FlowCellect Mouse™ Th1 Differentiation Tool Kit includes a CD3 Coated Activation Plate, complete Differentiation Media and Activator/Re-Stimulation Reagents. Detailed assay instructions are included to assist in three culture steps: Activation, Expansion and Re-Stimulation to complete Th1 cell generation in 6 days.

      Day 1=Activation: Culture CD4+ cells with growth factors and TCR co-stimulator on an anti-CD3 coated 96-well plate for 3 days.
      Day 4=Expansion: Expand activated/developing Th1 cells in a larger tissue culture plate.
      Day 6=Re-Stimulation: Differentiated Th1 cells are re-stimulated with mitogens to induce Th1 signature cytokine production on day 6.

      Background InformationCD4+ T cells play a central role in the function of the immune system. They help B cells produce antibodies, enhance and maintain responses of CD8+ T cells, regulate macrophage function, orchestrate immune responses against a wide variety of pathogenic microorganisms, and regulate immune responses.

      Effector T cells are derived from naïve CD4+ progenitor T cells, after maturational complex interactions with antigen-presenting cells in a permissive milieu, including antigenic type and load, and cytokine signaling. Committed CD4+ T cells can differentiate into four major lineages: Th1, Th2, Th17 and regulatory T cells with distinct cytokine products and biological functions.

      The primary role of the Th1 subtype is to address intracellular pathogens, by producing IFNgamma (IFNg), interleukin (IL) -2 and tumor necrosis factor alpha/beta to help cellular immunity. The differentiation to the Th1 lineage is promoted through a series of complex signaling events such as IL-12 which is mainly produced by macrophages as well as IFNg which produced by NK, CD8 and Th1 cells. In addition, neutralizing antibody-mediated IL-4 depletion enhances the generation of Th1 cells in vitro, suggesting that IL-4 which is signature cytokine for Th2 lineage, antagonizes Th1 polarization.
