      OverviewThe reagent is an anti-mouse Ig antibody, F(ab')2 fragment from sheep. The F(ab')2 fragments are obtained by pepsin digestion and conjugated with digoxigenin-N-hydroxy-succinimide ester and stabilized in buffer.

      PURIFICATION: The sheep-lgG is purified by ion exchange chromatography and the specific IgG was isolated by immunosorption. Cross-reacting antibodies were removed by further affinity chromatography using human-Ig and bovine Ig coupled to a solid phase.

      Application NotesThis highly specific reagent allows a reliable and sensitive detection of mouse- Ig, especially also in such situations where interference of immunoglobulins of other species than mouse might occur. The digoxigenin labeled antibody can be used analogous to the biotinylated antibody in a number of immunological detection systems (Savage 1986) such as ELISA (Baumgarten 1986; Kendal et al. 1983), immunohistochemistry, respectively immunocytochemistry (Volson 1984; Gonatas et al. 1987), immunoblotting (Gabridge et al. 1986; Ogata et al. 1983) and screening of monoclonal antibodies, whereby the various anti-digoxigenin conjugates allow a high flexibility.


      Immunohistochemistry: 20-40 μg/mL

      ELISA: 0.05-2 μg/mL

      Immunoblotting: 10-20 μg/mL

      The concentrated stock solution (200 μg/mL) should be diluted with PBS, pH 7.4 containing bovine serum albumin (BSA) 0.5% (w/v).

      Optimal working dilutions must be determined by the end user.
      Biological Information
      SpecificityThe reaction profile of the antibody was examined in an ELISA sandwich system, where the same antibody was used for coating and, labeled with digoxigenin, also for detection. According to this assay, the antibody reacts with mouse Ig classes IgA, IgG, IgM and the mouse IgG subclasses IgG1, IgG2a, IgG2b, IgG3 as well as with the l- and k- chain. Cross-reactivity was determined with normal sera, diluted 1:10, of various animal species. The antibody shows no cross-reactivity (<0.01%) with fetal calf serum (FCS) and normal serum from bovine, rabbit, goat, horse, and human. Weak reaction can be detected with normal serum from guinea pig (<0.1%). Cross-reactivity can be detected with normal serum from rat (<10%).
