      Overview Useful for measuring protein kinase A (PKA) or protein kinase C (PKC) activity in crude cell extracts or semi-purified enzyme preparations. The assay kit is based on an ELISA method which utilizes a synthetic PKA/PKC pseudosubstrate and a monoclonal antibody which recognizes the phosphorylated form of that peptide. May be used for screening inhibitors and activators of PKA and/or PKC. Supplied with a user protocol.

      Product Usage Statements
      Intended use The Calbiochem® Protein Kinase Assay Kit, Non-Radioactive is intended for the qualitative determination of cytosolic protein kinase C (PKC) and/or protein kinase A (PKA) using cell lysates or purified enzymes. The kit is suitable for screening of inhibitors and activators of PKA and/or PKC , monitoring the purification process for PKA or PKC, and measuring the effect of various pharmacological agents on PKC and/or PKA activity in biological systems. Please note that translocation of PKC from the cytosol to the membrane is a hallmark of PKC activation for most PKC isoforms. Since this kit measures cytosolic PKC rather than membrane-associated PKC, there may be a decrease in cytosolic PKC activity in cell samples that have been activated. The kit is not suitable for determination of calcium or phospholipid dependency with purified PKC.

      NOTE: Please review the entire protocol before commencing with the assay. The Protein Kinase Assay Kit, Non-Radioactive has been tested and optimized for measuring either PKC or PKA activity using cell lysates from FM3A cells.
