      Background InformationThe processing of the type 1 transmembrane protein beta-amyloid precursor protein (APP) by gamma secretase is thought to be a key event needed for the deposition of the beta-amyloid peptide in senile plaques found in Alzheimer's disease. The proteolytic activity of the gamma secretase cleaves the transmembrane domain of APP and releases the beta-amyloid peptide. Early or aberrant gamma-secretase cleavage of APP underlies the majority of early onset, familial Alzheimer's disease. {Guo, M et al (2003) Hum Mol Genet. 12(20):2669-78}. The gamma secretase consists of complex of four membrane associated proteins including presenilin, nicastrin, Aph-1 and Pen-2 {Gu, Y et al (2003) J Biol. Chem. 278(9):7374-7380; Kimberly, WT et al (2003) PNAS, USA 100(11):6382-6387}. It is believed that Aph-1 is a membrane protein that directly associates with presenilin and nicastrin within the gamma-secretase protein complex and stabilizes the presenilin within the complex, and allows the for proteolytic processing of presenilin and activation of the gamma-secretase activity{Takasugi, N et al (2003) Nature 422(6930):438-441}.
