      Background InformationTLR3 (Toll-like Receptor 3) is a member of the Toll-like receptor family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. They recognize pathogen-associated molecular patterns (PAMPs) that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. The various TLRs exhibit different patterns of expression. TLR3 is most abundantly expressed in placenta and pancreas, and is restricted to the dendritic subpopulation of leukocytes. TLR3 recognizes dsRNA associated with viral infection, and induces the activation of NF-kappaB and the production of type I interferons. TLR3 thus plays a role in host defense against viruses.

      SUBUNIT STRUCTURE: Interacts with MYD88 via their respective TIR domains. Interacts (via transmembrane domain) with UNC93B1. Interacts with TICAM1. Homodimer formation is triggered by ligand binding and is required for TLR3 signaling. Binding of double-stranded RNA is required for the interaction with SRC.

      SUBCELLULAR LOCATION: Endoplasmic reticulum membrane; Single-pass type I membrane protein. Endosome membrane.

      TISSUE SPECIFICITY: Expressed at high level in placenta and pancreas. Also detected in CD11c+ immature dendritic cells. Only expressed in dendritic cells and not in other leukocytes, including monocyte precursors. TLR3 is the TLR that is expressed most strongly in the brain, especially in astrocytes, glia, and neurons.
