      OverviewThe Light Diagnostics Mumps IFA Kit is intended for the culture confirmation of mumps virus in infected cells.

      Test Principal:

      The Light Diagnostics Mumps IFA Kit utilizes the indirect immunofluorescence assay for the detection of mumps virus. The assay utilizes a blend of monoclonal antibodies directed against mumps antigen. The antibodies will bind specifically to the mumps antigen present in the cells fixed in the substrate antigen wells. The antibodies are then detected with a secondary fluorescein isothiocyanate-(FITC) labeled goat anti-mouse antibody. An apple-green fluorescence is observed in the cytoplasm of mumps virus-infected cells.

      Summary and Explanation:

      Mumps began to be recognized in the fifth century by description of parotitis1. Bilateral or unilateral parotitis is the most common clinical feature of mumps. Meningitis, encephalitis, epididymoorchitis, oophoritis, polyarthritis, and pancreatitis commonly occur with mumps virus infection2. Mumps infection results in long term immunity but the disease is preventable though vaccination since vaccine introduction in 19673,4. Vaccination has resulted in a dramatic decrease in the incidence of reported mumps cases, but unvaccinated populations continue to sustain the virus5,6,7.

      The mumps virus belongs to the family Paramyxoviridae and is a member of the genus Paramyxovirus. It is an enveloped pleomorphic virus ranging in size from 100 to 600 nm1.

      The mumps virus can be grown in a variety of established cell lines8. Rhesus monkey kidney cells are most often used for the isolation of mumps virus from patient specimens9. Since not all strains of mumps virus produce cytopathic effects, cell culture confirmation is performed by immunofluorescence.

      • Mumps Monoclonal Antibody - (Catalog No. 5028). One (1) 2 mL vial containing mumps monoclonal antibody.
      • Mumps Control Slides - (Catalog No. 5029) Two (2) slides each containing one well of mumps virus infected cells (positive) and one well of non-infected cells (negative).
      • Anti-Mouse IgG / FITC Conjugate - (Catalog No. 5008). One (1) 10 mL vial containing labeled mouse IgG antibody with 0.02% Evans Blue counterstain.
      • PBS - (Catalog No. 5087). One (1) packet of phosphate buffered saline salts yield 1 liter when dissolved in distilled water. Store in a clean, closed container at room temperature.
      • Tween 20 / Sodium Azide Solution (100X) - (Catalog No. 5037). One (1) 10 ml vial containing polyoxyethylene sorbitan monolaurate (Tween 20) and sodium azide (NaN3) concentrate to be diluted 1:100 in PBS.
      • Mounting Fluid - (Catalog No. 5013). One (1) 10 mL dropper vial containing tris buffer, glycerin, fluorescence enhancer, and sodium azide as preservative. Store at room temperature.

      Storage ConditionsWhen stored at 2-8° C, the Mumps kit is stable up to the expiration date printed on the kit label. Do not freeze or expose to elevated temperatures. Discard any remaining reagents after the kit expiration date.

      Warnings And Precautions:

      · Sodium azide (NaN3) is used as a preservative in several components and is toxic if ingested. NaN3 may react with lead and copper plumbing to form highly explosive metal azides. Upon disposal, flush with large volumes of water to prevent build up in plumbing.

      · Pooling or alteration of any reagent may cause erroneous results.

      · Incubation times or termperatures other than those specified may give erroneous results. Any such change must be validated by the user.

      · Mounting Fluid contains a fluorescence enhancer that may be destructive to mucous membranes. Avoid direct skin or mucous membrane contact. If contact occurs, flush with large volumes of water.

      · Do not use reagents after expiration.

      · Do not allow slides to dry at any time during the staining procedure.

      · Handle all specimens and materials coming in contact with them with the same precautions as for potentially infectious material. Decontaminate with 0.05% sodium hypochlorite (a 1:100 dilution of household bleach).

      · Do not expose reagents to bright light during storage or incubation.

      · Avoid contact with Evans Blue (present in the anti mouse IgG / FITC conjugate) as it is a potential carcinogen. If skin contact occurs, flush with large volumes of water.

      · Do not mouth pipette reagents.

      · Do not substitute reagents from other manufacturers.

      · Alteration of protocol provided may cause erroneous results.

      · Avoid cross contamination between samples.
