The K-LISA™ PTK Screening Kit, an ELISA-based protein tyrosine kinase (PTK) activity assay, is a rapid, non-radioactive method to screen for PTK activity of purified enzyme samples as well as in cell lysates or subcellular fractions and tissue extracts. This colorimetric assay can detect activity at picomolar concentrations with purified PTKs and in as little as 5 µg total protein from cell lysates and 20 ng total protein from tissue extracts. The kit uses three different random polypeptide substrates (EAY, E4Y, or EY) that are composed of different molar ratios of glutamic acid (E), alanine (A), and tyrosine (Y), (1:1:1, 4:1, and 1:1, respectively) and are immobilized on two 96-well plates. To study general PTK activity, substrate selectivity, or changes in PTK activity associated with inhibitor or mutational effects by either comparative or ratiometric analysis.
