A pentapeptide designed from the central hydrophobic region in the N-terminal domain of Aβ that acts as a β-sheet breaker. Inhibits Aβ fibrillogenesis, disassembles preformed fibrils in vitro and prevents neuronal death induced by fibrils in cell culture. Significantly reduces Aβ deposition in vivo and completely blocks the formation of amyloid fibrils in rat brain. Binds to the monomeric/dimeric Aβ peptides with high affinity (Kd ~70 nM), inhibits peptide conformational changes and blocks fibril formation.
