The BacMagic™-3 Transfection Kit provides a positive selection for creating baculovirus recombinants in preparation for recombinant protein expression in insect cells. The kit improves on the traditional method for generating recombinant baculoviruses by eliminating the tedious, time-consuming steps of plaque purification. Derived from an AcNPV genome, BacMagic-3 DNA is deleted for a portion of the essential open reading frame (ORF) 1629, preventing nonrecombinant virus form replicating in insect cells. The nonessential chitinase (chiA) gene is deleted to improve production of secreted and membrane target proteins. Additionally, the v-cath gene, a cathespsin-like cysteine protease, is deleted resulting in significantly improved quality and yield for most target proteins. Unlike BacMagic™-2, the BacMagic-3 genome is deleted for an additional three viral genes (p10, p24, and p26). These genes are nonessential to virus in tissue culture and improve protein expression. The BacMagic-3 Transfection Kit contains the BacMagic-3 DNA, Insect GeneJuice® Transfection Reagent, and a positive control plasmid to monitor co-transfection efficiency. For optimal performance, Sf9 cells and BacVector® Medium are also included in the transfection kit.
