BLT5615rna carries an ampicillin-resistant plasmid that supplies large amounts of capsid protein 10A. Growth of T7Select10-3, T7Select1-1 and 1-2 clones on BLT5615rna requires the addition of IPTG to induce production of the 10A protein. BLT5615rna is a less inhibitory host than BLT5403, where 10A expression is controlled by a lacUV5 promoter.
Stability of RNA in T7 phage lysates prepared with different E. coli host strains
Radiolabeled U1hpII RNA (5 pmol) was incubated at room temperature with T7 phage lysates (5 × 109 pfu) prepared by growth on either E. coli BLT5615 or BLT5615rna, and samples were analyzed by gel electrophoresis. (Reproduced with permission from reference 2)
