A spectrophotometric assay kit based on the method described by Paglia and Valentine. Glutathione peroxidase (GPx) catalyzes the reduction of an organic peroxide while forming oxidized glutathione (GSSG). GSSG is reduced to GSH by the enzyme glutathione reductase, and NADPH is oxidized to NADP in the catalytic cycle. GPx activity is quantitated by measuring the change in absorbance at 340 nm caused by the oxidation of NADPH. Each kit can be used to perform up to 100 assays.
