71892  iFOLD® Protein Refolding System 3

The iFOLD® Protein Refolding System 3 is designed to screen for optimal refolding conditions for proteins expressed as inclusion bodies. The iFOLD System 3 utilizes FoldACE™ Reagents, which are novel, detergent-free refolding additives that have been demonstrated to significantly enhance refolding of certain proteins. The kit provides a systematic evaluation of 96 conditions representing different buffering systems, pH values, redox agents, and 5 different FoldACE Reagents. Following cell lysis with the aid of Lysonase™ Bioprocessing Reagent, membrane components and contaminating proteins trapped within inclusion body pellets are removed by a series of washes with Triton X-100. The purified inclusion bodies are reduced and denatured by addition of Tris(2-carboxyethyl) phosphine (TCEP) and guanidine hydrochloride and refolded by rapid dilution into the iFOLD System refolding buffers. Components for cell lysis, inclusion body washes and denaturation, and a pre-dispensed 96-well plate-based matrix of refolding buffers are included in the kit. Each kit is sufficient for screening up to 96 refolding conditions (91 experimental and 5 buffer-only control wells) for a single protein.
