The pCITE®-4-a-c(+) vectors are designed for dramatic enhancement of the efficiency of in vitro translation of cloned sequences. The maps for pCITE-4b(+) and pCITE-4c(+) are the same as pCITE-4a(+) with the following exceptions: pCITE-4b(+) is a 3702 bp plasmid; subtract 1bp from each site beyond Nco I at 610. pCITE-4c(+) is a 3704bp plasmid; EcoR V cuts at 618. Add 1bp to each site beyond EcoR V (e.g. BamH I cuts at 625). The f1 origin is oriented so that infection with helper phage will produce virions containing single-stranded DNA that corresponds to the T7 RNA polymerase coding strand. Therefore, single-stranded sequencing should be performed using the T7 terminator primer.
