The pIE1-3 vector is designed for constitutive expression of recombinant proteins from the baculovirus ie1 promoter in stably-transformed insect cells (1). The plasmid contains all promoter sequences known to be important for ie1-mediated gene expression in uninfected insect cells as well as the hr5 enhancer element. pIE1-3 lacks an ie1-driven translation initiation site; therefore, the ATG start codon must be supplied by the insert. To obtain stably-transformed cells, a pIE1-3 recombinant is cotransfected with pIE1-neo, which contains the Neo selectable marker under ie1 control.
