The Prussian blue reaction describes the reaction of any free ionic iron (Fe3+) in the tissue with potassium hexacyanoferrate (II) aqueous hydrochloric acid solution. The reaction product is a typical bright blue particle that precipitates as a non-soluble complex in blood, bone marrow or tissue cells, thus, detecting free cellular iron. The reaction is used, for instance, to diagnose hemochromatosis (liver) and asbestosis (lung and joints) in histological approaches and bone marrow diseases in hematology. In tissue sections, amounts of down to 0.0002 µg ionic iron can be detected.
