Application Comments
For use as a negative control, preincubate 1 µg of peptide with 1 µg of antibody for 1 h at room temperature. The epitope is unique for AQP3 and is not present in other known related proteins. It is highly homologous in mouse (17/18 identical) and human (15/18 identical) AQP3 protein. Recognition of AQP3 can be fully inhibited with the soluble AQP3275-292 blocking peptide. Variables associated with assay conditions will dictate the optimal working dilution.

Reconstitute the antibody with 50 µl deionized water to make a 300 µg/ml stock solution. Further dilutions should be made in a physiological buffer containing a carrier protein such as 1-3% BSA. Avoid freeze/thaw cycles of solutions. To reduce nonspecific staining, centrifuge all antibody preparations before use (10000 x g for 5 min) to remove any aggregates. Reconstitue the peptide with 100 µl deionized water, aliquot, and freeze (-20°C).

Immunoaffinity purified rabbit polyclonal antibody. Recognizes the ~40 kDa and ~29 kDa aquaporin-3 proteins. Supplied with a control peptide (immunogen).
Background
AQP3 may function as a water and urea exit mechanism in antidiuresis in collecting duct cells. It is also slightly permeable to urea and glycerol. AQP3 may play an important role in gastrointestinal tract water transport and in glycerol metabolism. AQP3 has a wide tissue distribution. In the kidney it is predominantly expressed in the inner medulla.
