      Background InformationGlutathione S-transferase class-mu 26 kDa isozyme (UniProt: P08515; also known as EC:2.5.1.18, GST 26, Sj26 antigen, SjGST). The glutathione S-transferases (GSTs) are a group of inducible enzymes that are important in the detoxication of many different xenobiotics in mammals. The GSTs achieve detoxication by catalyzing the conjugation of reduced glutathione (GSH) to various electrophilic substrates. GSH functions as a cellular storage pool of reduced thiols. GSH conjugation is the first step in mercapturic acid synthesis which aids in the protection of the cell by enhancing the excretion of toxic metabolites. Mammalian cytosolic GSTs are classified into four classes, termed as , , , and . These classes differ in their tissue-specific expression and distribution within tissues. They exist as homo-or heterodimeric combinations of subunits. These classes of GST isozymes differ in their specificity toward xenobiotic or endogenous substrates. GST from Schistosoma japonicum (SjGST) is reported to operate via a sequential bisubstrate catalytic mechanism. It displays a hybridization of and class features. Its structure consists of a 4-stranded β-sheet and 3 -helices in domain 1 and 5 -helices in domain 2. It possesses relatively limited selenium-independent glutathione peroxidase activities but is able to catalyze the glutathione conjugation of members of the trans,trans-alka-2,4-dienal, trans-alk-2-enal and 4-hydroxyalk-2-enal series of reactive carbonyls, the secondary products of lipid peroxidation. It participates in the immunogenicity to the vertebrate host and have been suggested as a potential component of a vaccine against schistosomiasis. It also serves as a useful tool in the development of a gene expression system that allows the fusion of a protein or protein fragment to the C-terminus of the enzyme. (Ref.: Lim, K., et al. (1994).       Entrez Gene SummaryCytosolic and membrane-bound forms of glutathione S-transferase are encoded by two distinct supergene families. At present, eight distinct classes of the soluble cytoplasmic mammalian glutathione S-transferases have been identified: alpha, kappa, mu, omega, pi, sigma, theta and zeta. This gene encodes a glutathione S-transferase that belongs to the mu class. The mu class of enzymes functions in the detoxification of electrophilic compounds, including carcinogens, therapeutic drugs, environmental toxins and products of oxidative stress, by conjugation with glutathione. The genes encoding the mu class of enzymes are organized in a gene cluster on chromosome 1p13.3 and are known to be highly polymorphic. These genetic variations can change an individual's susceptibility to carcinogens and toxins as well as affect the toxicity and efficacy of certain drugs. Null mutations of this class mu gene have been linked with an increase in a number of cancers, likely due to an increased susceptibility to environmental toxins and carcinogens. Multiple protein isoforms are encoded by transcript variants of this gene.
