      Background InformationATP-dependent DNA helicase PIF1 (EC 3.6.4.12; UniProt Q9H611; also known as DNA repair and recombination helicase PIF1, Petite integration frequency 1, PIF1 5'-to-3' DNA helicase homolog, PIF1/RRM3 DNA helicase-like protein) is encoded by the PIF1 (also known as PIF, C15orf20) gene (Gene ID 80119) in human. Human PIF1 helicase and the yeast ortholog share common substrates in vitro, exhibiting specificity toward telomeric DNA, synthetic stalled DNA replication fork-like structures, especially G4-DNA. PIF1 functions to maintain undisrupted DNA replication fork progression during normal cycling conditions and to support resumption of DNA synthesis after prolonged S-phase arrest. Cancer cells are reported to exhibit increased dependency on PIF for DNA replication. PIF1-depletion results in a combination of apoptosis, reduced survival, and hypersensitivity to therapeutic DNA replication inhibitors in several cancer cell lines independent of p53 status.
