Ret is a receptor tyrosine kinase that mediates the activation of several signaling pathways including the PLC gamma, Ras, JNK and inositol phosphate pathways. Ret mutations have been shown to be causative in several diseases, including Hirschsprung's disease (HD), papillary thyroid carcinoma, multiple endocrine neoplasia (MEN) 2A, MEN 2B, and familial medullary thyroid carcinoma. Ret is initially produced with a signal peptide sequence (a.a. 1-28), the removal of which yields the mature Ret (a.a. 20-1114). Ret can be posttranslationally cleaved into the extracellular cell-membrane-anchored RET cadherin 120 kDa fragment (a.a. 29-707) and the soluble RET kinase fragment (a.a. 708-1017).
