Background Information	GBP5, also known as GBP-TA antigen, or GTP-binding protein 5, or Guanine nucleotide-binding protein 5 or GBP-5, and encoded by the gene GBP5, is a protein that plays a role in the inflammatory response to pathogens and may also play a role in apoptosis. GBP5 stimulates cryopryrin (NLRP3 protein) assembly of the intracellular inflammasome, a multiprotein oligomer consisting of caspase 1, PYCARD, NALP and sometimes caspase 5. Inflammasomes act as sensory complexes that alert the immune system to the presence of infection or tissue damage. Nucleosomes and GBP5 are expressed in myeloid cells and are a component of the innate immune system; GBP5 acts as key rheostat for activation and assembly of the inflammasomes. Additionally, GBP5 is a GTPase that homodimerizes upon GTP-binding; dimerization is required for the second hydrolysis step from GDP to GMP. GBP5 is localized to the cytoplasm and intracellular membranes via its lipid-anchor sequences.

GBP5 promotes NLRP3 inflammasome assembly and immunity in mammals. Shenoy, Avinash R, et al. Science, 336: 481-5 (2012) 2012 Show Abstract Inflammasomes are sensory complexes that alert the immune system to the presence of infection or tissue damage. These complexes assemble NLR (nucleotide binding and oligomerization, leucine-rich repeat) or ALR (absent in melanoma 2-like receptor) proteins to activate caspase-1 cleavage and interleukin (IL)-1β/IL-18 secretion. Here, we identified a non-NLR/ALR human protein that stimulates inflammasome assembly: guanylate binding protein 5 (GBP5). GBP5 promoted selective NLRP3 inflammasome responses to pathogenic bacteria and soluble but not crystalline inflammasome priming agents. Generation of Gbp5(-/-) mice revealed pronounced caspase-1 and IL-1β/IL-18 cleavage defects in vitro and impaired host defense and Nlrp3-dependent inflammatory responses in vivo. Thus, GBP5 serves as a unique rheostat for NLRP3 inflammasome activation and extends our understanding of the inflammasome complex beyond its core machinery.	22461501
