Background InformationInterleukin-12 subunit alpha (UniProt: P43431; also known as IL-12A, Cytotoxic lymphocyte maturation factor 35 kDa subunit, CLMF p35, IL-12 subunit p35) is encoded by the Il12a gene (Gene ID: 16159) in murine species. IL-12 is a heterodimeric pro-inflammatory cytokine composed of covalently linked p35 and p40 subunits. The two genes encoding p35 and p40 are unrelated and located on separate chromosomes. However, both genes encoding IL-12 need to be expressed coordinately in the same cells to produce the biologically active heterodimer. It has been reported that in the absence of p35 subunit p40 is secreted as a monomer or a homodimer; however, p35 subunit is secreted only when associated with p40. IL-12 is secreted by activated monocytes, macrophages, and dendritic cells, and has been shown to target naïve, resting CD4+ T cells to promote their proliferation and secretion of cytokines. Microbial products are reported to be strong inducers of IL-12 by these cells. Production of IL-12 is shown to be regulated by positive and negative regulatory mechanisms. IL-10 is considered as a potent inhibitor of IL-12 production and acts by blocking transcription of both of its encoding genes. TGF-β is also an inhibitor of IL-12 production. It acts by reducing the stability of IL-12 p40 mRNA. IL-12 can act as a growth factor for activated T and NK cells and can enhance the lytic activity of NK/lymphokine-activated killer cells. It also induces the production of interferon-gamma and favors the differentiation of T helper 1 (Th1) cells and forms a link between innate resistance and adaptive immunity. (Ref.: Trinchieri, G.
