  • PAX transactivation activation domain-interacting protein
  • protein encoded by CAG trinucleotide repeats

Background InformationPAX transactivation activation domain-interacting protein (PTIP) is a ubiquitous nuclear protein composed of 5 tandem BRCA1 carboxyl-terminal (BRCT) domains. PTIP was initially identified as an interacting, transactivation protein that binds to the C-terminal end of Pax2. Since its discovery, studies have shown PTIP to bind several Pax proteins, playing a critical role in response to DNA damage. It also has activity as a phosphoserine/ phosphothreonine-specific binding module, and can discern ATM and ATR gamma irradiation induced phosphorylated substrates. PTIP is involved in early cellular development and is likely to have a role in cell proliferation as well.

Entrez Gene Number
Entrez Gene SummaryThis gene is a member of the paired box (PAX) gene family and encodes a nuclear protein with six BRCT (breast cancer carboxy-terminal) domains. This protein plays a critical role in maintaining genome stability, condensation of chromatin and progression through mitosis. [provided by RefSeq].

UniProt Number
UniProt SummaryFUNCTION: Functions as a key component of the DNA damage response. Functions as phosphoserine or phosphothreonine-specific binding modules. Recognizes substrates phosphorylated by the kinases ataxia telangiectasia-mutated (ATM), ataxia telangiectasia and RAD3-related (ATR) in response to gamma irradiation. May play a role in PAX2-dependent transcriptional regulation. Plays a role in early development, most probably in cell proliferation.

SUBUNIT STRUCTURE: Interacts with the C-terminal transactivation domain of PAX2 (By similarity). Interacts with C16orf53/PA1; the interaction is direct. Component of the MLL3/MLL4 complex, at least composed of MLL3, MLL4, ASH2L, RBBP5, DPY30, WDR5, NCOA6, KDM6A, PAXIP1/PTIP and C16orf53/PA1. Some fraction of the complex contains MLL2 instead of MLL4 and is named MLL2/MLL3 complex.

SUBCELLULAR LOCATION: Nucleus membrane; Peripheral membrane protein; Matrix side (By similarity).

DOMAIN: The BRCT domains function as a single module and are necessary and sufficient for phospho-specific binding.

SEQUENCE SIMILARITIES: Contains 5 BRCT domains.

SEQUENCE CAUTION: The sequence AAB91434.1 differs from that shown. Reason: Frameshift at positions 609 and 628.

The sequence AAP21865.1 differs from that shown. Reason: Erroneous gene model prediction.
Molecular Weight~120 kDa observed.
Other isoforms may be observed in some lysates.
