Background InformationThyrotropin receptor (UniProt: P16473; also known as Thyroid-stimulating hormone receptor, TSH-R) is encoded by the TSHR (also known as LGR3) gene (Gene ID: 7253) in human. TSH-R is a multi-pass membrane protein of the G-protein coupled receptor (GPCR) family that is synthesized with a signal peptide (aa 1-20), which is subsequently cleaved off to generate the mature form that contains four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. The extracellular domain is linked to a distal signal-specific domain the hinge region that is attached to a transmembrane domain consisting of extracellular and intracellular loops. It serves as a receptor the thyroid-stimulating hormone and TSH binding to this receptor induces thyrocyte growth and proliferation primarily by activating the adenylate cyclase signaling pathway. Clone CS-17 acts allosterically and serves as an inverse agonist for TSH-R constitutive activity. When administered in vivo it is shown to suppress thyroid function and reduces serum T4 levels in BALB/c mice. (Ref.: Chen, RR., et al. (2018). Thyroid. 28(77); 933-940; Chen, CR., et al. (2007).
