Bead-based flow cytometric assays enable sensitive, precise quantification of analytes within a sample. When directed toward proteins or peptides, such assays are essentially ELISAs on a bead. Samples are combined with fluorescently labeled microparticles (beads) covalently conjugated to a capture antibody. Analytes captured on the beads are identified with detection antibodies and a fluorescent label. A major advantage of bead-based assays over traditional protein quantification methods (such as ELISA) is the capacity for multiplexing, as bead-based assays allow simultaneous quantification of multiple analytes in a small sample volume.

Panel Overview

Three subunits of Complex IV (cytochrome c oxidase), which is the last enzyme complex of the mitochondrial respiratory chain, are encoded in the mitochondrion, two of the three mitochondrial DNA encoded proteins play a catalytic role. The enzyme is altered in Alzheimer's disease, and there are reports of reduced amounts of this complex in cancer cells when these cells become hypoxic. The proteins of Complex II (succinate ubiqinone oxidoreductase) are entirely encoded in the nucleus and serves as a reference in the BeadPlex® Human Mitochondrial Toxicity: Mitochondrial Biogenesis Panel.

Examples of inhibitors of mitochondrial biogenesis are the nucleoside analog antiviral drugs designed to interfere with the reverse transcription of invading viral RNA, but with the side effect of inhibiting mitochondrial DNA replication. A second and well-established mode of toxicity affecting mitochondrial biogenesis is drug-induced interference with mitochondrial protein synthesis. The preferential binding and subsequent inhibition of mitochondrial ribosome is believed to account for the dose-limiting mitochondrial toxicity associated with the antibacterial drug therapies such as chloramphenicol. Both classes of drugs cause the reduction of complex IV quantity by inhibiting either mitochondrial DNA replication or mitochondrial protein sysnthesis, whereas complex II, which is entirely encoded by nucleus and synthesized by cytosolic ribosome is not affected. Therefore the ratio of complex IV to complex II quantity serves as a good indicator for mitochondrial biogenesis.

The BeadPlex Human Mitochondrial Toxicity: Mitochondrial Biogenesis Panel is designed for the in vitro quantitative determination of Complex II and IV in cell lysates and tissue extracts, and subsequently allows end-users to determine whether mitochondrial biogenesis has been up-regulated or down-regulated as a result of drug interactions or conditions such as oxidative stress and mitochondrial genetic disease.
