General description
Protein jagged-2 (UniProt: Q9QYE5; also known as Jagged2) is encoded by the Jag2 gene (Gene ID:16450) in mouse. The Notch signaling pathway is an intercellular signaling mechanism that is essential for proper embryonic development. Members of the Notch protein family are transmembrane receptors that are critical for various cell fate decisions. Jagged2 is one of several ligands that activate Notch and related receptors. It is a single-pass type I membrane protein and is synthesized with a signal peptide sequence (aa 1-23), the removal of which yields the mature Jagged2 with a large extracellular domain (aa 24-1082), a transmembrane domain (aa 1083-1103) and, a cytoplasmic domain (aa 1104-1247). Jagged2 expression varies across different tissues and developmental stages and is found to be highest in fetal thymus, epidermis, foregut dorsal root ganglia and inner ear. In 2-weeK-old mice its abundantly found in heart, lung, thymus, skeletal muscle, brain and testis. Its expression overlaps partially with Notch1 expression. Jagged2 has been implicated in various diseases, mainly due to its role in the Notch signaling pathway. The interaction of Natural Killer (NK) cells with dendritic cells (DC) contributes to NK cell-mediated antitumor or antimicrobial responses. Studies show an involvement of Jagged2 Notch interaction in augmenting NK cell cytotoxicity mediated by dendritic cells and indicated that, the Jagged2 Notch axis plays a crucial role in DC-mediated NK cell cytotoxicity. It is also suggested that manipulation of this interaction may provide an approach to induce potent tumor immunity or to inhibit certain autoimmune diseases caused by NK cell activation. This clone functions as an agonist by promoting Treg cell differentiation. Aberrant Jagged2 expression can lead to uncontrolled cell proliferation and tumorigenesis in cancers such as breast cancer, lung cancer, and glioblastoma. (Ref.: Kijima, M., et al. (2008). Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
