Hard-to-Filter Samples in Dissolution Testing

Often times dissolution testing samples are viscous and/or contain high levels of particulates. Viscous samples can greatly slow down filtration rate, and high levels of particulates may clog the filter membrane’s pores. For these dissolution testing samples, we recommend using filtration devices optimized for hard-to-filter samples.

High Particle Filtration

25 mm HPF Millex® syringe filters are the right choice for high particulate load samples. HPF Millex® syringe filters include a graduated glass fiber prefilter to remove larger particles and either a 0.20 μm or 0.45 μm membrane filter for fine filtration. This combination of prefilter and membrane provides significantly greater throughput than standard filters without prefiltration media, especially when filtering particle-laden solutions. HPF Millex® syringe filters are available in bulk for individual sample filtration and in tubes for use with automated filter changing systems.

Samplicity® G2 Filtration System for Hard-to-Filter Samples

Manually filtering hard-to-filter HPLC samples can require excessive manual force, greater time per sample, and lower recovery—all reducing laboratory efficiency. In contrast, the vacuum-driven Samplicity® G2 filtration system filters even highly viscous samples in seconds, with minimal manual force.

Millex Samplicity® filters with 0.45 µm PVDF membrane were used to efficiently clarify hard-to-filter pharmaceutical samples.

Force required for traditional syringe filtration versus Samplicity® system.

1.0 mL samples of 1% Pepto-Bismol were filtered by 15 users, either through hydrophilic polytetrafluoroethylene (PTFE) Millex® syringe filters with a 10 mL syringe, or through 0.45 µm hydrophilic PTFE Millex Samplicity® membrane filters using the Samplicity® filtration system.
