Eukaryotes have the ability to recognize and degrade transcripts that harbor premature signals for translational termination. Nonsense and frameshift mutations that result in these premature termination codons play a role in many genetic diseases and some cancers. Nonsense-mediated nRNA decay (NMD) is mediated by several factors including rent1/hUpf1, rent2/hUpf2, and hUpf3. When assembled, these proteins form the surveillence complex. Disruption of the mouse Rent1 gene has been reported to completely disrupt NMD and result in embryos that re-absorb shortly after implantation.
