      Applications
      Key Applications Immunoblotting (Western Blotting)
      Application NotesImmunoblotting (1:2000-1:5000)
Application CommentsThe appearance of the smaller bands is concurrent with a reduction in the amount of the procaspase by immunoblot analysis. A weak, unrelated, ~30 kDa band may be detected in some samples. Antibody works only weakly in paraffin-embedded tissue sections. Reactivity with the yeast homolog is weak. Variables associated with assay conditions will dictate the optimal working dilution.

      ApplicationImmunoblotting (1:2000-1:5000)
      Application Data
Detection of human caspase-3 by immunoblotting. Samples: Whole cell lysates from HeLa cells (lane 1), Jurkat cells (lane 2), and Jurkat cells treated with staurosporine (lane 3). Primary antibody: Anti-Caspase-3 Rabbit pAb (Cat. No. 235412) (1:1000). Detection: chemiluminescence.
DescriptionImmunoaffinity purified rabbit polyclonal antibody. Recognizes the ~34 kDa latent and the ~20 kDa and ~18 kDa active forms of caspase-3.
BackgroundCaspase-3 is a member of the group II family of interleukin-1β convertases (ICE) cell death effector (CED) cysteine proteases. Caspase-3 and caspase-6 are the major active caspases in apoptotic cells, suggestion that activation of caspase-3 is a pivotal event in the execution phase of apoptosis. Upon activation, procaspase-3 is cleaved into a large p17-p20 subunit (depending on the activator) and a smaller p12 subunit.
