      Description
      OverviewA cell-permeable thienopyridone compound that acts as a direct, potent, selective, reversible, and allosteric activator of AMPK (EC50 = 0.8 µM, 1.9 µM and 2.2 µM in partially purified AMPK from rat liver, muscle and heart, respectively; 0.7 µM for AMPKα1,β1,γ1 isoform and 1.1 µM in HEK cells). Acts by inhibiting the dephosphorylation of AMPKT172. Does neither affect GPPase and FBPase activities at 100 µM, and a panel of 67-receptors and ion-channels at 10 µM nor alter the intracellular levels of ATP and AMP. Specifically stimulates hepatic AMPK in ob/ob mouse model (30 mg/kg, i.p.), improves insulin-sensitivity and regulates glucose and lipid metabolism. Shown to reduce mRNA expression levels of several gluconeogenic enzymes.
