      Background InformationPotassium channel subfamily K member 9 (UniProt: Q9NPC2; also known as Acid-sensitive potassium channel protein TASK-3, TWIK-related acid-sensitive K(+) channel 3, Two pore potassium channel KT3.2, Two pore K(+) channel KT3.2, KCNK9) is encoded by the KCNK9 (also known as TASK3) gene (Gene ID: 51305) in human. KCNK9 is a homodimeric multi-pass membrane protein of the two-pore domain potassium channel that maintains resting membrane potential of the cell. It is highly sensitive to changes in pH. It can also form heterodimer with KCNK1. It is found mainly in cerebellum, but has also been detected in adrenal gland, kidney, and lung. Over-expression of KCNK9 has been reported in several carcinomas, including breast, lung, colon, and prostate. It is reported to confer resistance to growth factor deprivation and hypoxia, which promotes tumor cell survival in hypoxic areas of solid tumors. KCNK9 has three cytoplasmic domains (aa 1-8, 129-158, and 240-374), four transmembrane domains, four extracellular domains, and two pore-forming domains (aa 89-101 and 195-207). Clone Y4 displays highest affinity for KCNK9 (KD = 0.7 nM) and induces channel internalization. Addition of Y4 to KCNK9-expressing carcinoma cells is shown to reduce their viability and increase cell death. It also inhibits the growth of human lung cancer xenografts and murine breast cancer metastasis in mice. (Ref.: Sun, H., et al. (2016). Nat. Commun. 7; 10339).
