      DescriptionFull length, recombinant, human TGF-β2 expressed as a chimeric protein (112 amino acids) containing the human TGF-β1 signal and latency associated peptide sequences with the mature TGF-β2 sequence. For use in growth inhibition assays and other assays designed to study cellular responses and receptor interactions involving TGF-β2. Expressed in NSO mouse myeloma cells and purified from conditioned medium. The mature protein is disulfide linked and has a predicted molecular weight of ~25 kDa.

      TGF β is prototypic of a superfamily of proteins with diverse activities. The TGF β proteins and other members of the family show growth, differentiation, and morphogenic activity in a wide range of biological systems being especially involved in many processes of tissue development and repair. Structurally, the TGF β1, β2, and β3 proteins are homodimers composed of disulfide linked chains 112 amino acids in length. Composition of TGF β proteins from different species is highly conserved at the DNA level as well as at the protein level showing in general >85% identity within a subtype and 64-82% identity between subtypes. TGF β1 and β2 were first identified on the basis of their biological activity and subsequently cloned from expression libraries. Human TGF β3 was identified first at the cDNA level and of the three major isoforms, appears to be the most biologically active. The expression of TGF β3 at various stages of morphogenesis and at different sites suggests an important role in the developmental process. Product should be titrated for optimal use in individual systems.

      Recommended reaction conditionsLyophilized protein must be reconstituted with 50 µl of 10 mM HCl/10% ethanol using siliconized, sterile pipet tips. Working dilutions should be made in sterile medium containing 1-10% serum or 0.1-1% BSA and used immediately, do not store working solutions.
