Background InformationArylsulfatase B (EC 3.1.6.12; UniProt P15848; also known as ASB, G4S, N-acetylgalactosamine-4-sulfatase) is encoded by the ARSB (also known as MPS6) gene (Gene ID 411) in human. Arylsulfatases consistute a group of enzymes found in the lysosomes of the liver, pancreas, and kidneys. Arylsulfatase B mediates the breakdown of two glycosaminoglycans (GAGs), dermatan sulfate and chondroitin sulfate. Arylsulfatase B-catalyzed removal of sulfate groups from chondroitin-4-sulfate (C4S) leads to the degradation of C4S. Cellular arylsulfatase B expression is also reported to be found in non-lysosomal compartments, including the plasma membrane. Arylsulfatase B is initially produced with a signal peptide sequence (a.a. 1-36), the removal of which yields the mature enzyme. Arylsulfatase B-deficiency due to ARSB mutations results in an accumulation of GAGs in lysosomes, which in turn can lead to Maroteaux–Lamy syndrome or mucopolysaccharidosis VI (MPS6).
