General description

Citrullination or deimination is the post-translational modification of arginine residues to citrulline residues in proteins. This process is catalyzed by enzymes called peptidylarginine deiminases (PADs).
The Anti-Citrulline (Modified) Detection Kit provides reagents for the modification and detection of citrulline-containing proteins by Western blot analysis. Modification of citrulline residues is created by a chemical reaction with 2,3-butanedione monoxime and antipyrine in a strong acid solution. Detection of the modified citrulline proteins uses a standard immunoblot protocol with an anti-Citrulline (modified) Human monoclonal antibody and a goat anti-Human IgG secondary antibody horseradish peroxidase (HRP) conjugate. This type of citrulline modification ensures the detection of citrulline residues in proteins regardless of neighboring amino acid sequences.

Preparation Note

Store Anti-Modified Citrulline monoclonal antibody at 2-8°C. Store Goat Anti-Human IgG, HRP Conjugate, Reagent B, and 0.5% FeCl3 at -20°C for 4 months from date of receipt.

Anti-Modified Citrulline, clone C4 (Part number MABS487). One vial containing 100 µL of Protein A purified recombinant human monoclonal IgG1λ in PBS containing 0.05% sodium azide. Goat Anti-Human IgG, HRP Conjugate: (Part number CS216591). One vial containing 50 µL of Goat anti-Human IgG conjugated to horseradish peroxidase (HRP) in PBS, pH 7.6, with 7.5 mg/mL BSA and diluted with StabilZyme® and glycerol to 50%. Reagent B: (Part number 20-253). One bottle containing 50 mL of 0.5% 2,3-butanedione monoxime, 0.25% antipyrine, 0.5 M acetic acid. 0.5% FeCl3: (Part number 20-255). One bottle containing 4 mL of 0.5% FeCl3. Frozen solution.
