Gene Information

human ... ANGPTL3(27329)

General description

The protein encoded by this gene is a member of the angiopoietin-like family of secreted factors. It is predominantly expressed in the liver, and has the characteristic structure of angiopoietins, consisting of a signal peptide, N-Terminal coiled coil domain and the C terminal Fibrinogen (FBN)-like domain. The FBN-like domain in angiopoietin-like 3 protein was shown to bind alpha 5 beta 3 integrins, and this binding induced endothelial cell adhesion and migration. This protein may also play a role in the regulation of angiogenesis. Angptl3 also acts as dual inhibitor of lipoprotein lipase (LPL) and endothelial lipase (EL), and increases plasma triglyceride and HDL cholesterol in rodents. Angptl3 inhibit endothelial lipase to catalyze HDL-phospholipid and increase HDL-PL levels. Circulating PL-rich HDL particles have high cholesterol efflux abilities. Defects in Angptl3 are the cause of familial hypobetalipoproteinemia type 2 (FHBL2) also called combined hypobetalipoproteinemia familial.
~70 kDa observed. The calculated molecular weight of this protein is 54 kDa, but can be observed at ~70 kDa due to high glycosylation. (Shimizugawa, t., et al. (2002). Journal of Biological Chemistry. 277:33742-33748.)

Immunogen

GST-tagged recombinant protein corresponding to the coiled coil region of mouse Angptl3.

Application

Detect Angptl3 using this Anti-Angptl3 Antibody validated for use in Western Blotting, Immunofluorescence, IHC(P).
Immunohistochemistry Analysis: A 1:100-500 dilution from a representative lot detected Angptl3 in human liver, human cardiac myocytes, mouse liver, rat liver, and rat kidney tissues.

Biochem/physiol Actions

This antibody recognizes the coiled coil region of Angptl3.

Analysis Note

Evaluated by Western Blot in HepG2 cell lysate.

Western Blot Analysis: A 1:1,000 dilution of this antibody detected Angptl3 in 10 µg of HepG2 cell lysate.

Obesity impacts the endocrine and metabolic functions of the adipose tissue. There is increasing interest in the role of epigenetic factors in obesity and its impact on diabetes and dyslipidemia. One such substance, miR-181, reduces plasma triglyceride levels in mice
