General description
Grainyhead-like protein 3 homolog (UniProt Q8TE85; also known as Sister of mammalian grainyhead, Transcription factor CP2-like 4) is encoded by the GRHL3 (also known as SOM, TFCP2L4, VWS2) gene (Gene ID 57822) in human. Mammalian grainyhead-like (GRHL1, GRHL2, and GRHL3) genes code for transcription factors and are named after their drosophila homolog grainyhead (GRH). These factors show tissue- and developmental stage-dependent expression patterns. Grainyhead-like 3 (GRHL3) is reported to play a protective role against squamous cell carcinoma (SCC) of the skin. Tumor suppressor phosphatase and tensin homolog (PTEN) is a direct target of GRHL3, Grhl3-knockdown in mice causes skin tumor formation as a result of reduced Pten expression. Consistently, downregulated GRHL3 and PTEN levels are found in human SCC skin samples. In addition, GRHL3 is a positive regulator of E-cadherin expression during mesenchymal epithelial transition (MET) in non-tumorigenic mammary gland cells, as well as in breast and lung cancer cells. High GRHL3 expression is found in early stages of breast cancer and the expression is greatly reduced in advanced stages of breast cancer.
