General description

Wnt/Wingless signaling controls many fundamental processeses, such as cellular growth and fate, and is known to be involved in embryonic segmentation, limb development and CNS patterning. Wnt signaling is also shown to influence postembryonic regulation of stem cell numbers and stem cell differentiation in epithelia, such as those of the colon and skin.
Wnt transduction is mediated by the association of Beta-Catenin with nuclear TCF DNA binding factors, including legless (lgs) and pygopus (pygo). Whereas lgs enclodes BCL9, pygo encodes a Pygopus (Pygo), a PHD finger protein. It is reported that BCL9 acts as an adaptor protein that recruits and physically links Pygo to the Beta-Catenin-TCF Complex. It is demonstrated that the recruitment of Pygo is required for this respective complex to function as a transcriptional co-activator of specific target genes.
