General description

GST-tagged recombinant mouse RAD18 C-terminal fragment.
~60/70 kDa observed. Given the difference of about 10 kDa in the molecular mass of the two
proteins, we concluded that the upper band was derived from monoubiquitination of the lower band (Miyase, S., et al. (2005). J. Biol. Chem. 280:515-524). Uncharacterized band(s) may appear in some lysates.

Application

Epitope: C-terminal region.

Biochem/physiol Actions

This polyclonal antiserum specifically detected RAD18 expression in wild-type, but not Rad18-/-, mouse embryonic fibroblasts (MEFs) by Western blotting and fluorescent immunocytochemistry (Tateishi, S., et al. (2003). Mol. Cell. Biol. 23(2):474-481).

Physical form

Rabbit polyclonal serum with 0.02% sodium azide.

Preparation Note

Stable for 1 year at -20°C from date of receipt.Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Evaluated by Western Blotting in MEF-1 cell lysate.

Western Blotting Analysis: A 1:500 dilutiion of this antibody detected RAD18 in 10 µg of MEF-1 cell lysate.
