Analysis Note

Evaluated by Western Blotting in mouse embryonic stem cell lysate.

Western Blotting Analysis: A 1:500 dilution of this antibody detected STAT3 trimethylated on Lys180 in 10 µg of mouse embryonic stem cell lysate.

Application

Immunoprecipitation Analysis: 5.0 µg from a representative lot immunoprecipitated STAT3 trimethylated on Lys180 in 0.5 mg of mouse embryonic stem cell lysate.
Peptide Inhibition Assay: Target specificity of a representative lot was confirmed by immunogen peptide blocking prior to Western blotting analysis (0.5 µg/mL) ) of mouse embryonic stem cell lysate (10 µg/lane) . Only the immunogen peptide, but not the corresponding peptide without the Lys180 trimethylation modification, blocked the detection of the target band.

General description

Signal transducer and activator of transcription 3 (UniProt P40763; also known as Acute-phase response factor, DNA-binding protein APRF) is encoded by the STAT3 (also known as ADMIO, APRF, HIES) gene (Gene ID 6774) in human. STAT proteins are latent cytoplasmic transcription factors that have the dual function of signal transduction and activation of transcription. STATs are activated by tyrosine phosphorylation in response to different ligands, after which they translocate to the cell nucleus. The N-terminal region is highly homologous among the STAT proteins and surrounds a completely conserved arginine residue. STATs are a part of the JAK-STAT signaling pathway – a major pathway of the immune system. All cytokines transduce critical signals through this pathway. STAT3 is known to be activated by IFN alpha but not IFN beta. The transcription factors associated with STAT3 are cJun and cyclic AMP responsive enhancer binding protein (CREB). STAT3 gene knockout in mice is lethal at the early embryonic stage.

A major focus of breast cancer research is to understand the mechanisms responsible for disease progression and drug resistance. Toward that end, it has been found that approximately two thirds of all human breast carcinomas overexpress the Estrogen Receptor α (ERα) protein and it remains the primary pharmacological target for endocrine therapy1,2. The normal cellular function of ERα is as a transcription factor that mediates a wide variety of physiological processes, many of which are dependent upon phosphorylation of the receptor at specific amino acid residues3,4. Indeed, ERα is known to be phosphorylated at a multitude of different sites, yet how these all correlate to disease remains unclear5. Here, we interrogated multiple sites of ERα for phosphorylation status by screening an extensive panel of different breast cancer patient samples and other non-breast cancer tissue microarray (TMA) slide samples to determine their relevance to disease.
