Anti-SLC4A11 Antibody

Evaluated by Western Blotting in mouse eye tissue lysate.

Western Blotting Analysis: 5.0 µg/mL of this antibody detected SLC4A11 in 10 µg of mouse eye tissue lysate.

Immunocytochemistry Analysis: A 1:80 dilution from a representative lot detected the expression of exogenously transfected human SLC4A11 in PS120 Chinese hamster lung fibroblasts (Courtesy of Professor Joseph A. Bonanno, O.D., PhD, Indiana University, Bloomington, IN).
Western Blotting Analysis: A representative lot detected SLC4A11 in cultured human and bovine corneal endothelial cell lysates, as well as in isolated rabbit and bovine corneal endothelium extracts (Jalimarada, S.S., et al. (2013). Invest. Ophthalmol. Vis. Sci. 54(6):4330-4340).
Western Blotting Analysis: A representative lot detected siRNA-mediated SLC4A11 downregulation in cultured bovine corneal endothelial cells (Jalimarada, S.S., et al. (2013). Invest. Ophthalmol. Vis. Sci. 54(6):4330-4340).
Immunocytochemistry Analysis: A representative lot detected predominant lateral membrane localization of SLC4A11 in isolated rabbit corneal endothelium, while peripheral staining was evident in cultured bovine corneal endothelial cells, by fluorescent immunocytochemistry staining of 2% paraformaldehyde-fixed, 0.2 % saponin-permeabilized cells (Jalimarada, S.S., et al. This Anti-SLC4A11 Antibody is validated for use in Western Blotting, Immunocytochemistry for the detection of SLC4A11.

Specificity of this polyclonal antibody was verified by antigen peptide blocking of target band detection in Western blotting application (Jalimarada, S.S., et al. (2013). Invest. Ophthalmol. Vis. Sci. 54(6):4330-4340). Epitope sequence is conserved between murine and human (UniProt A2AJN7 and Q8NBS3) and present in all four human SLC4A11 spliced isoforms reported by UniProt.

Sodium bicarbonate transporter-like protein 11 (UniProt A2AJN7; also known as Bicarbonate transporter-related protein 1, NaBC1, Sodium borate cotransporter 1, Solute carrier family 4 member 11) is encoded by the Slc4a11 (also known as BTR1) gene (Gene ID 269356) in murine species. Bicarbonate (HCO3-) transporters are the principal regulators of pH in cells and play a vital role in acid-base movement in various tissues. There exist four types of HCO3- transporters, namely Cl-/-HCO3- exchangers, Na+/HCO3- cotransporters (NBCs), a K+/HCO3- cotransporter, and a Na+-driven Cl-/-HCO3- exchanger. SLC4A11 is a ubiquitously expressed 14-transmembrane protein existing as dimers within the plasma membrane. Originally classified as bicarbonate transporter, later stiudy conducted with cultured primary bovine corneal endothelial cells (BCECs) indicated that SLC4A11 mediates Na+-coupled hydroxide (OH-) transport, but not borate (B(OH)4-) or bicarbonate (HCO3-) transport in corneal endothelium. SLC4A11 gene mutations in human are linked to endothelial dystrophies, including CDPD (Corneal dystrophy and perceptive deafness), CHED2 (Corneal dystrophy, endothelial 2, autosomal recessive), and FECD4 (Corneal dystrophy, Fuchs endothelial, 4).

~110 kDa observed. Target band size appears larger than the calculated molecular weights of 96.75 kDa (mouse) and 99.58/45.76/98.18/103.1 kDa (human isoform 1/2/3/4) due to glycosylation. Uncharacterized band(s) may appear in some lysates.
