General description

NUB1 is a critical control protein for the ubiquitin proteasome system in that it regulates NEDD8 activity by specifically targeting NEDD8, ubiquitin D (FAT10), and their conjugates to the proteasome for degradation. There are two isoforms, isoform one, (NUB1L) is more efficient at NEDD8 recruitment than isoform 2. Interestingly NUB1 is localized primarily in the nucleus of resting cells and its expression is wide spread from leukocytes in the blood to liver cells and even skeletal muscle cells. Also interesting is that NUB1 was identified early as an auto-antigen in renal carcinomas. NUB1 appears to operate via binding to Asn-51 on NEDD8 which complexes with the UFD1-NPL4 complex that facilitates degradation via the proteasome.
