General description
Charged multivesicular body protein 6 (UniProt: Q96FZ7; also known as Chromatin-modifying protein 6, Vacuolar protein sorting-associated protein 20, Vps20, hVps20, CHMP6) is encoded by the CHMP6 (also known as VPS20) gene (Gene ID: 79643) in human. CHMP6 is a human orthologue of yeast Vps (vacuolar protein sorting) 20, a component of ESCRT (endosomal sorting complex required for transport)-III. Various CHMP6 orthologues in organisms ranging from yeast to humans contain the N-myristoylation consensus sequence at each N-terminus. CHMP6 is an ubiquitously expressed protein that has an acidic C-terminus and the basic N-terminus that render the protein in a closed, soluble and inactive conformation through an auto-inhibitory intramolecular interaction. The open and active conformation, which enables membrane binding and oligomerization, is achieved by interaction with other cellular binding partners, probably including other ESCRT components. CHMP6 is probable core component of the endosomal sorting required for transport complex III (ESCRT-III), which is involved in multivesicular bodies (MVBs) formation and sorting of endosomal cargo proteins into MVBs. MVBs contain intraluminal vesicles that are generated by invagination and scission from the limiting membrane of the endosome and mostly are delivered to lysosomes enabling degradation of membrane proteins.
