Application

Immunohistochemistry: 1:250-1:2,500 using free floating sections by the PAP technique on rat cholinergic areas. Tissues should be fixed with a mixture of nitrobenzyl alcohol and gluteraldehyde (Chagnaud, 1989). Gluteraldehyde is required for staining; not recommended for formalin or paraformaldehyde fixations without gluteraldehyde.

ELISA: 1:1,000-1:5,000

Immunocytochemistry: 1:1,000-1:5,000

Western Blot: 1:1,000-1:2,000

Optimal working dilutions must be determined by end user.

This Anti-Acetylcholine Antibody is validated for use in IH for the detection of Acetylcholine.

Biochem/physiol Actions

Acetylcholine. By immunohistochemistry on rat basal forbrain and spinal chord sections, the antibody specifically stains nerve cell bodies and fibres (Chagnaud, 1989).

The cross-reactivities were determined using an ELISA test by competition experiments with the following compounds:

Compound Cross-reactivity

Choline-GA-BSA 1

Choline-AA-BSA 1/536

Choline-SA-BSA 1/2,500

Farnesol-BSA 1/>50,000

Retinoic acid-BSA 1/>50,000

Myristic acid-BSA 1/>50,000

Acetylcholine 1/>50,000

Choline 1/>50,000

Abbreviations:

(GA) Glutaric anhydride

(AA) Adipic acid

(SA) Succinic acid

(BSA) Bovine Serum Albumin
